Effect of milk supplementation on bone growth in pre-pubertal pigs.

Achieving peak bone mass during childhood and adolescence is associated with a decrease in the risk of osteoporosis and osteoporotic related fractures later in life. Milk has always been associated with increased bone health, strength, and development, but assessing the direct effect of milk on multiple properties of bone development is difficult with a human model. Therefore, this study aimed to use a pig model to evaluate the effect of milk consumption on the growing skeleton. For this, we used 24 pre-pubertal pigs fed a normal growing diet supplemented with 750 mL of whole milk or an isocaloric maltodextrin solution for 12 wk. The experiment was run in 2 trials: trial 1 utilized 4 male and 2 female 5-wk-old Yorkshire pigs per group while trial 2 utilized 7-wk-old Duroc × Yorkshire pigs of 6 males per group. Ultrasonography was used throughout the trial to record in vivo bone growth. After 12 wk, the pigs were euthanized and bones of the appendicular skeleton and the mandible were collected. All bones were analyzed with dual energy x-ray absorptiometry (DEXA). A 3-point bending test was used for biomechanical testing. After fracture, the cortical bone thickness was taken at 3 regions of the femur. Data were analyzed using GLM procedure of SAS with TRT and Experiment (and Sex for Trial 1) as fixed effect and pig as random. To evaluate differences between groups, a Student’s t-test was used. Significance was declared with P < 0.05. Of the DEXA measurements taken, only the bone mineral density of the mandible was significantly lower in pigs receiving milk (P = 0.04). Upon further evaluation, the difference was in the mandibular body (P = 0.03). There was no difference in maximum force, stiffness, or extension of the femur from the biomechanical tests. The medial cortical thickness of the femur was larger in the control group (4.4 ± 0.1 mm vs. 4.0 ± 0.4 mm, P = 0.03), while the lateral thickness was larger in the group receiving milk (7.4 ± 0.8 mm vs 6.2 ± 0.9 mm, P = 0.05). Short-term milk supplementation did not increase bone mineral density, growth, and strength. The biological significance and the link between milk and the larger lateral cortical thickness in the femur and the lower bone density in the mandible remain unclear.
